چکیده

In a recent publication1 we reported on the isolation and identification of flavonoids and cinnamic acid derivatives from spinach chloroplast preparations. These cinnamic acid derivatives and flavonoids are obtained as low molecular fractions I and II from a water extract — called S^eth — from ether treated lyophilized chloroplasts, which is assumed to contain functional components of the photosynthetic electron transport chain between photosystem I and ferredoxin 2 '3 . We have discussed the possibility that a phenolic compound might be the prosthetic group of this functional component (possibly the primary acceptor) and also the relation of our phenolic compounds to other fractions of various activity isolated from chloroplasts by other authors 1. We have shown that the chromophoric group of low molecular fraction I consists of two esters of p-coumaric acid 1. The chemical nature of the compounds esterified to p-coumaric acid remained to be elucidated. We wish to report here on their identification. The p-coumaric acid derivatives of low molecular fraction I, absorbing at 312 nm, were obtained from the water extract of ether treated lyophilized chloroplasts as previously described After alkaline hydrolysis of its chromophore, the resultant alkaline solution was passed through a Dowex 50 WX 8 column and the p-coumaric acid extracted with ether1. The compound, which was originally esterified to p-coumaric acid, could now be found in the water phase. It was identified as meso-tartaric acid by paper chro-

Cytochrome c reducing substances in photosynthetic electron transport.

Photosynthetic cytochrome c reduction by isolated chloroplasts is mediated by substances with characteristics of a “Cytochrome c Reducing Substance” (CRS). A water soluble extract of ether treated, lyophilized chloroplasts (called SL-eth), related to the primary acceptor complex of photo system I, has CRS-activity. It contains p-coumaroyl-meso-tartaric acid. By comparison with model substances...

Fat Metabolism in Higher Plants: LVII. A Comparison of Fatty Acid-Synthesizing Enzymes in Chloroplasts Isolated from Mature and Immature Leaves of Spinach.

Chloroplasts isolated from immature leaves of spinach (Spinacia oleracea) differ in enzyme levels from those isolated from mature leaves. On a chlorophyll basis, immature chloroplast preparations had 5- to 6-fold higher capacity to synthesize fatty acids from 2-(14)C-acetate compared to plastids isolated from mature leaves. This difference was correlated with higher activities for the enzymes, ...

Lipid and Fatty Acid composition of chloroplast envelope membranes from species with differing net photosynthesis.

Lipid and fatty acid compositions were determined for chloroplast envelope membranes isolated from spinach (Spinacia oleracea L.), sunflower (Helianthus annuus L.), and maize (Zea mays L.) leaves. The lipid composition was similar in sunflower, spinach, and undifferentiated maize chloroplast envelope membranes and different in maize mesophyll chloroplast envelope membranes. The predominant lipi...

Sequence of the genes for tRNACys and tRNAAsp from spinach chloroplasts.

We have determined the map location and primary structure of two fragments of spinach chloroplast DNA which encompass the genes for tRNACysGCA and tRNAAspGUC. Identification of the genes for these two RNA species is based on the sequence of their anticodon triplets and on a comparison of the sequences with those of the equivalent tRNAs from Escherichia coli. Each gene occurs only once on the sp...
